Il 14 giugno 1800, giorno della battaglia di Marengo, il patriota lombardo Cesare Angelotti evade dal carcere di Castel Sant'Angelo e si rifugia, travestito da donna, nella chiesa di Sant'Andrea, dove il pittore Mario Cavaradossi, di malcelate tendenze giacobine, sta lavorando a un affresco. Costui, che è l'amante di Floria Tosca, cantante famosa, nasconde Angelotti in una casa di campagna. Sfruttando abilmente la gelosia di Tosca, il barone Scarpia, reggente della polizia pontificia, scopre il rifugio del patriota, che per non consegnarsi ai gendarmi si uccide. Per averlo protetto, Cavaradossi è arrestato e condannato a morte. Sentendosi colpevole, Tosca, rinchiusa anche lei a Castel Sant'Angelo, sta per concedersi a Scarpia in cambio della vita dell'amato, ma, ottenuto un salvacondotto per due, e l'assicurazione che Mario sarà fucilato per finta, uccide il barone. Cavaradossi viene invece fucilato sul serio, per cui Tosca, disperata, si getta dagli spalti del castello.
