Siamo nel XXIII secolo. Un "intruso", distrutte agevolmente le astronavi di difesa di Klington, si sta dirigendo verso la Terra. Il Capitano James T. Kirk, nonostante il lungo periodo di permanenza alla base, assume la direzione del veicolo spaziale "Uss Enterprise", con disappunto di Willard Decker che viene costretto a fare da secondo. Il veicolo, tra l'altro, non è del tutto a punto. Comunque delle 431 persone dell'equipaggio, il motorista Scott, il timoniere Sulu, l'addetto alle comunicazioni Uhura, l'ufficiale di controllo Chekov, la dottoressa Christine Chapel, l'addetto al carico Janice Rand, la calva pilota Ilia fanno ciascuno il proprio dovere
